    I've entered the OpenDNS server addresses in the manual network setup, but should I disable DNS relay on my D-Link router (DIR-615)?

    Well, you don't really need it, but it shouldn't cause problems. It is just a local resolver cache in the router, somewhat like the local resolver cache in a computer OS. Either can be disabled or enabled with no harm done.

    If you have set up blocking categories with OpenDNS, and you have visited sites that you now want blocked, you need to flush those caches. On the router, that may be a disable/re-enable, or there may be a flush option. Also, flush the browser cache. http://www.opendns.com/support/article/209
